The role of oral bacteria in producing nitric oxide, which is beneficial for heart and mental health, is less well known.
Many types of oral bacteria convert inorganic vegetable nitrate into nitrite. Nitrite is a precursor to nitric oxide, a signature molecule that enlarges blood vessels and sends messages between neurons in the brain.
In older people, reduced availability of nitric oxide can contribute to hypertension (high blood pressure) and decreased brain function.
New research shows that drinking beetroot juice, rich in inorganic nitrate, can alter the balance of bacteria in the mouth by favoring species that promote better mental and heart health.
The findings of this study help to explain the proven benefits of beetroot juice for lowering blood pressure in people with hypertension.
Other vegetables that are high in nitrates, including cabbage, lettuce, spinach and legumes, can provide similar benefits.
Scientists at the University of Exeter in the United Kingdom led the study, which was published in the journal Redox Biology.
“We are really excited about this finding, which has a significant impact on healthy aging,” said lead author Prof. Anni Vanhatalo.
He adds: “Keeping this oral microbiome healthy in the future can reduce the negative changes in blood vessels and the aging process associated with aging,” he adds.
The study included 26 people aged 70–80.
Investigators occasionally assigned these participants to one of two groups. While asking both groups to drink 140 milliliters of beetroot juice a day for ten days, the experimental group drank nitrate-rich juice, while the control group drank nitrate-depleted juice.
The aroma and taste of the two drinks were similar.
After a 3-day bathing period, groups changed, and participants drank another drink for 10 days.
Investigators asked participants not to use mouthwash during the study.
Before, during, and after the test, they provide a battery for physical and cognitive exercises. They also identified genetic variants in the saliva samples from the participants.
Compared with the placebo regimen, subjects who drank regular beetroot juice had higher numbers of oral infections related to good cardiovascular health.
Their saliva also contains low numbers of germs associated with inflammation.
On average, the systolic participants’ blood pressure dropped by 5 millimeters of mercury after 10 days of drinking nitrate-rich beetroot juice. In addition, studies have shown high levels of nitric oxide in their blood.
Those who drank nitrate-rich juice also performed better on a continuous attention test.
Researchers used a statistical method to identify two different clusters of bacteria that thrived in the mouths of participants when they drank common beetroot juice.
These two collections or “modules” (labeled MM5 and MM6) were not only associated with the same mouth but were also associated with improved mental functioning and blood pressure, respectively.
The authors suggest that these nitrate-sensitive oral bacteria may make probiotic appropriate to correct age-related decline in cardiovascular health.
A third group of animal species, a previous study that has been associated with inflammation, failed to grow in the mouths of participants who drank regular beetroot juice.
The collection included a variety of pathogens, including Clostridium kengoku.
Professor Vanhatalo warns that more research will be needed to determine if these findings apply to other groups of people.
“Our participants were healthy, active, older people with normal blood pressure,” he explains.
“Dietary nitrate has reduced their blood pressure to an average, and we are keen to find that the same can be said of other races and people with poor health,” he adds.
The authors also note that the process they used to identify bacteria, called 16S ribosomal sequining, determines the number of organisms rather than total value or metabolic activity.
The final amount will influence the magnitude of nitrite production in the mouth and, therefore, the availability of nitric oxide.
In addition, the authors wrote that the 16S sequence (which successfully reads a single gene sequence) is not very good at identifying certain types of viruses.
To develop probiotics, they say, researchers will need to use a process called genome sequencing, which is more expensive but more accurate.
